Whole thing starts with end of "Betrayal ol killian qatar" mission.
I'm fairly sure your progress through the missions has no influence on the game starting or not starting... it's more likely due to external factors like driver updates.
I'd suggest fiddling with the compatibility mode, putting it to different things and see if that helps. And make sure you run in administrator mode :-\
Have you disabled the Origin In-Game thing? It's also known to cause problems.
