Ooo, tough choice! I have all of those and think they're all great additions. They all come with good questlines, are voiced, and have some fun visable tasks.
Jasper gets bonus points from me at the moment because he can craft pills in SH, and pills are needed more than anything else now I'm closing in on the higher levels. If you have them in your inventory, his Stonecutters and Tapball skins also have their own short questlines.
Disco Stu also helps in SH with yoga mats although they have never felt like a bottleneck. Love his audio clips and he has a Stonecutters outfit too. I especially love the character so for me it's neck-and-neck between him and Jasper.
Duffman is iconic and I bought him before Jasper or Disco Stu. I think all his tasks are visible but apart from 1 they're all the same, they just happen outside different buildings. He's had the least involvement in the game/events so far, so he'd be my close 3rd choice.
Duffman because I like the building most, and the game releases so many related items that go with it.
Also, if you have the Steel Mill that came with Roscoe, duffman dances with Roscoe and Smithers and Roscoe there. Though if you don't particularly like duffman dance, it can be tiresome. (Plus his 6-hour "promote duff" task on his Segway looking thing is one of my favorites)
He has a neat task over the ballpark too if you've purchased/had that.
He really only has like 1 or 2 indoor tasks, really one of the few characters with this flexibility (I think Kang is close to this as well).
It really depends on preference.
-If simply picking for your favorite on the show - that's up to you
-If picking to assist with the game, Jasper probably has the most use (used in SH for pills (greater need than Stus yoga mats)). Was used in Tapball and Stonecutters events.
-If picking for task quality, I think Jasper is most diverse with visuals, but Duffman has more visual tasks (Stu has few visible tasks)
-If picking based on building, Duffman/Brewery wins for it's flexible design, multiple game items with Duff- this building goes on Squidport too.
Really Stu is unfortunately the worst of the 3, despite being a fan favorite on the show.

That is my advice as well.
