Try to run Origin and the "MassEffect2.exe" as Administrator.
Do you use any Mods?
Did you edited one of the games Ini files?
If that don't helps, create a DxDiag in text file format and post it with your next reply. You can do that with the "choose file" button at the bottom right corner of the reply window.

I'm afraid running both Origin and the MassEffect2.exe as admin doesn't change anything.
I used the "Mass Effect Updated/Improved Textures Mod" for my second Shep in ME1, I can't recall changing any ini files though..

There are several "Update;taskhostw" errors in your DxDiag, do you get a error message at system start?
Run the Windows System File Checker Sfc /scannnow and post the concluding message.
Furthermore, your Win 10 is at build 14393. The Creators Update should have been installed by now, I assume that this is because of the above error, or did you blocked the update manually?

I've encountered no error messages so far. Though I must confess I'm not all that educated when it comes to this kind of stuff..so if there's nothing obviously wrong I probably won't notice it.
Also, I can't say I know anything about a Creators Update. I don't usually block updates.
The System File Checker didn't find any "Integritätsverletzungen", if that helps you in any way..

OK, lets get your system up to date first.
Go here, the website should tell you: "Wir haben festgestellt, dass Sie Windows 10 nutzen. Wenn Sie auf die neueste Version aktualisieren möchten, klicken Sie auf Jetzt aktualisieren"
Just do that.
(Note: As usual you should back up your important data before you preform a major update.
