Try to run Origin as an Administrator.

@jessliv If that doesn't work, the save folder also might accidentally be set to read-only. It's located in C:\Users\YourUsername\Documents\BioWare\Mass Effect 3. Right-click on the Save folder and click Properties, then make sure near the bottom of the dialog in the Attributes section that Read-only is not ticked (untick & click Apply if it is). Then run the game and see if saving now works.
