No. You have not lost any Ice Shroom seeds.

The reward at 500 crowns is 80 seeds.  If you add up all the seeds earned up to this point, you should have noticed it matches the amount of 250 seeds required to unlock the plant.

Once you unlock the plant, the amount of 250 seeds is consumed.
