Have you checked in the audio options to see if the mic is turned off? If you hit ESC and go to options and choose the audio tab it should look something like this

From there you can click on "VOIP" and choose either "off", "push to talk" or "always on". Make sure you're not set to off. If you want people to be able to hear everything you say put it on always on, otherwise push to talk means you just have to push a button to let people hear what you're saying - it's set to Left ALT as default. If you want to change this you'll need to go into customize controls:
