I don't think this would work in multiplayer; the bosses are severely over-powered by comparison to the average character and factoring them into multiplayer team play would lead to all sorts of balancing issues in a game which already seems to be a chore to balance.
Now, as a special Co-Op mode I think it has promise. You spawn as a Boss character and have to fend off 10 waves of increasingly more numerous and powerful enemies with the option of three other players joining as regular character classes as support.
You wouldn't have as many balancing concerns because any other players in the game would be on your team and the opposition would be all AI.
