Yes, you can.

You will still need to login into Origin but you can do so in offline mode.

I would activate the game, then play for 5 min. Then exit. Then set game to offline mode, then start the game.

If that all works, you can remove the Internet connection and play.

You will need to connect to update the game if you want to have bug fixes and new content and for that you'll also need Internet.
