Well, if we are doing characters that appeared only once, how about Rev. Lovejoy's Daughter (or was it his niece?)

The bad girl that Bart fell in love with could give his charcter alot to do in the game.

I really want Marvin Monroe, EA will probably release his memorial hospital at some point but I want the character. He's have to have his annoying voice though- its staple to his character. Plus he didn't die due to actor death, the writers don't like him though
