The Murder Knife, which is required to obtain the Assassin specialization, should be available to craft at the requisition table in Skyhold once you collect all the ingredients (the one supervised by Morris).
Could you try to repair option in Origin to see if it helps? Eventually, try to load a different save if you have one.
