You play a Origin copy? If yes, first try to repair the game (in Origin > my games > right click ME1 > Repair Game)
If that don't work, please report back

Yep, I'm on an Origin version and I already tried to repear the game several times. Unfortunately, it's getting stuck at the middle of the repearing...- @Mwinie
Du you have an Mods running, or did you changed any of the ini files?
If not, the only way I see is a complete new installation of the game.
Deinstall ME1 from the Windows control panel, reboot, and install the game again.
