@jakash284 I'm not sure do I understand but you can only have 100% boost from every specialization. If you have other gambling buildings close then there's your answer.

The total specialisation boost will have been replaced by other Gambling buildings when you moved it, when you moved it back it will have provided the "top up" boost it could over and above the buildings already there... total population is the best measure, not per building boost.
There is no need to wait for sims to move back in, it is instant.
