There a couple of ways to deal with the issue you ran into.

The first way is to prevent dust from building up on an inaccessible room.  I use the lowest level basement on all my residential lots as a storage room where I stick stuff like the Christmas tree.  There are no stairs or ladders to it.  When I first got Bust the Dust I noticed that dust and the dust bunnies started showing up there.  Not a big deal until I found that the dust piles and dust bunnies blocked me putting stuff in the basement.  I never got any of the Filth Fiends though.  And I didn't know they will set fires until reading your post.

Anyway, I have found a way to keep that basement clean so I don't get the dust piles and bunnies.  But you have to have the Cat and Dogs Expansion pack.  That pack has two robot vacuums.  So you get one, do all the upgrades on the robot and the base (they are really 2 separate objects and each part has to be upgraded separately), set it to run on auto every six hours and put it in your room.  I put one on every floor of my houses now and don't have any dust buildup at all.  They do break on occasion so you do need to check that they are still running every now and then.  But my house now stays pristine, including the inaccessible basement.

But you say you only have the Bust the Dust pack so that might not be an option for you so you will need to get a sim into the space to clean every so often.  There are two ways to do that and they allow you to teleport a sim there.  If you had Spa Day, there is a teleport ability once you level the Wellness skill up to level 7 or 8.  So that might not be an option for you either since you don't say you have that pack.

The other way is with a cheat.  Turn testing cheats on.  Then right click on a spot in the room where you want to go.  You should see a teleport option.  Use that to get a sim into the room to clean (have to have a vacuum in the sim's inventory) or to put out the fire.

This happened to me just last night and why I'm here now!

There was a "room" at the top of my roof that was there strictly for a glass roof top to sit on (picture doesn't have it in because I removed after the incident). There is no way to access this "room", it's just for decoration!

I got a notification that a fire had started "nearby" - I looked everywhere and found NO fire, so I dismissed it. Eventually the fire spread and started to burn the entire roof. I noticed as I leveled up to the top, that the "room" where the fire started was overwhelmed by dust and bunnies. I could not edit the lot because, fire. I couldn't get up there because, no stairs.

Luckily my sims could teleport and I had them teleport to the roof. Unfortunately I had one sim teleport to the room and one to the roof. That sim in the room started to burn before they could get the fire under control. The commands for "put self out" didn't come up until after I hit the "debug (reset)" on my sim and waited a few minutes.

I did manage to get it under control and removed the "room" because I didn't want to have to watch for another fire or clean that extra room since my sims are already cleaning three floors of this house manually. I do have the robot vacuum on the main floor, didn't think about throwing one up there in the Spare Oom.

It would really help if we just had a cheat to shift click on ground to get rid of dust. I'd be more happy with not spawning dust bunnies at all on inaccessible areas but I'd figure it would be easier to implement this cheat.

Even better would be that dust bunnies would only set on fire such things that would make sense catching on fire. Like a fridge, they can actually be a fire hazard if you never vacuum behind it. And no amount of dust will spontaneously ignite without anything to spark the fire so empty concrete rooms just shouldn't cause fires. With sims logic perhaps washers/dryers, stoves, stereos and generators could also catch on fire in addition to fridges. 

Dust bunnies will eventually cause fires which means we'll have fires in inaccessible areas that will go on until all of the concrete floor has burned. Just like real life firefighters, sims should also be able to extinguish a fire burning inside a roof instead of just standing still and eventually leaving.
