I believe genetics is determined at conception. I had a couple give birth to a daughter and I wanted her to look a bit more like dad so I kept downloading them to different saves while still pregnant to see if I can get different features to appear with each birth and that didn't work. So I assume when they are already pregnant, genetics is defined. I like my game as realistic as possible so I don't allow my same-sex couples to have genetic children with both providing the genetics (I really like the adoption feature for this); however, I did accidentally delete a dad once to a single mom but I still think his genetics showed up. He was an unknown entity in the daughter's family tree.
