No; alliances cannot be made or broken in-game. They are set up in advance in the lobby.

Ok, cool. Thank you very much for getting back to me. How do you set them up in the lobby?

You'll find this in the Drop-Down menu on the side of your Player Slot.
Where it says Team 1 by Default for the Hosting Player. Next to where you select House Colour.
