Made some tests.
Weapon: Revenant
Hipfired and walking and Taking fire from Enemys.
Chars with Toughness:
Turian Havok and Ghost:
- no penalty when firing weapon
- penalty walking and taking fire from enemys
- it seems that with taking a pack wich restores the shields they are not slowered from enemy fire
N7 Destroy:
- no penalty
Batarians all:
- no penalty
Krogans all:
- no penalty
Asari Justikar:
- penaltyTurian Sentinal and Soldier:
- penaltyChars without Toughness :
- slowered by bothMost of the chars with toughness having Damage resistance except the Turian Soldier.
Most of the chars with toughness having more Shields and health than other Chars.
The Asari Justikar has damage resistance from reave but not so much shields and health.
Other char for exaple Salarian and the N7 Sentinel having temporarly DR from enery drain if skilled and the N7 Sentinel has more Shields but a little bit lesser health than the Asari Justikar.
Also the Quarians (female) have equal Shields or more (male),
Conclusion:
I thought toughness could be related to walking and hipfiring a weapon like the revenant and getting not slowered and also not getting slowered by walking and taking fire from Enemies.
Maybe the Turians and the Asari Justikar should get the same ability like the Krogans and Batarians.
I hope you see my point.

Maybe an Developer can give a good explanation what thouness realy means.

"What is Toughness/Melee anyway?
Well Krogan has toughness/melee and Turian only has toughness.
I'm going with toughness being a higher default weight capacity or health"I agree with ajhere, the toughness means that you take less damage from your enemies, more resistant compare to others characters.
Build yourself a Krogan Vanguard
Level Up to give him maximum Biotic Charge, Barrier, Battlemaster, & Rage
Arm him with an N7 Pirahna
Have fun !! Run around the map charging, headbutting, & roaring with laughter.
This is a good example of "toughness"
