Maybe a dumb question but i will ask anyway...

I am on level 20 with 112,000 sims living there, i don't see any of these super-skyscappers in my city.

Do i need to do anything to obtain them?

Do i need to add more of the other specialization building?

My city looks pretty built up, but i still do not have any one them.

A building must have coverage from at least two different specializations when it upgrades to top tier (the +1.8k population upgrade) for it to become one of the super-skyscrapers listed. Your existing top-tier buildings cannot become super-skyscrapers (aside from bulldozing and rebuilding).
