Vader if not lead is one of the worst Sith from my experience, under EP lead he's okay but under Zaul sucks and don't see much greatness under Nihilus. Dooku with his counter constantly gaining HP back is a solid choice I would say and if Sidious Zeta unique applies after Nihilus lead he will have 80+k health, definitely a solid toons to include.
Sith Assassin works great under Zaul as a JE-like booster, don't know how well she'd do under Nihilus but probably not a bad choice, Trooper looks like a nice option as well because of his tankiness.
I know you loose out on EP stun but EP isn't very tanky and could be taken out quickly, same counts for Maul who is a slug when not leading, and if he doesn't get to move he can't gain all that extra max health, also his unique health gain after 3rd enemy death is useless with Nihilus as leader, keep that in mind.
If you run a Darth Nihilus lead you need to approach the matter a bit different than with Zader or Zaul or even EP, his leader ability benefits toons that attack frequently or have a good pool of protection to begin with, that's why I say no to Maul and EP.

I will say I have one difference in opinion on the Vader comment. I run an EP(l) with a maxed out Vader and he is one of my best arena characters outside of Zylo. The key with vader is he needs support to maximize the effectiveness of his Force Crush and Culling attack. Outside of Dooku with his counter and then getting another turn, not many characters get 3 attacks in a row. Being able to knock out a powerful attacker like Biggs, EP, TFP and others and then ability block works very well.
IMO, he needs B2 to mow down and Boba helps prevent any cleanses or tenacity up from Rex.
I'm not sure how that translates to a DN lead, but I can tell you after facing several Zaul teams that he makes short work of Sith that don't have a lot of health/protection.
