To sum up all info we have gathered:
to find Big Claw, either send homer (and big claw) on their 3 hour task to the nature reserve. When you collect from Homer, Big Claw will also be right there to tap. (Credit to johnde68) I prefer to use the method of going to build mode and hiding everything except characters.

You can only play the minigame every 24 hours, Big Claw will flash red if the 24 hours has not yet elapsed. I play the game when I collect from my wailing walls and launch the rocket on a morning so I know it will be ready to play. Another idea would be to synch with the time you do friends visits if you do them daily.

When you tap him the first time after 24 hours, he will have a different animation and claw the ground, as shown here:

Tapping it 3 times a day yields 50XP and either $250, $500 or 2 Pharmaceutical each time. Even if your Pharmaceuticals are full they will still be added. Any Pharmaceuticals dropped will also count towards any daily challenges too.
