Thanks, sorry that the regular steps aren't doing the job.
I want to try from a different angle, can you go to C:\Program Files (x86)\Origin Games\Mass Effect 2\__Installer\vc\vc2005\redist and run the application there as an administrator?
No help either.
I also manually uninstalled this program from my computer and installed it again from that folder. Did a repair also.

Please make sure that the automatic update funktion in Origin is enabled. In Origin > Origin (in the menu bar) > Application Settings > Automagically keep my games up to date.

Yeah those have been selected. I've also tested with beta participation.

You mean with the Origin beta client?

Yeah. I thought I'd give it a go.
No special updates have appeared though.

@spI4t Good thinking.
This is a "nasty" issue, because it's hard to get to the bottom of this error. In the most cases I saw the error is solved as soon as Origin updates the game.
Please go to your ">drive>:\Program Files (x86)\Origin Games\Mass Effect 2\Binaries" folder start the "MassEffect2Config.exe" and look up the version number of the ME2 exe under "game".
Also try to disconnect your Internet connection, close Origin completely and start the Game from the ME2 EXE directly.

Alright.
MassEffect2.exe: v1.02
MassEffect2Launcher.exe: Empty
MassEffect2Config.exe: v1.03
Uninstall Mass Effect 2.exe: Empty
When I disconnected and started from MassEffect2.exe directly there comes a "Activate Your Game" window and it asks my email or Origin credentials. Of course I can't login when disconnected. When connected it gives the error again when I log in.
I tried creating a new Windows profile to my PC with admin privileges but this didn't work either. Same error.

That the MassEffect2Launcher.exe is empty is strange.
What version of Windows are you running? The Windows account you are using is a Administrator account?
You do restart the PC sometimes, not just put it into hibernation?
Another possible solution is to deinstall ME2 again, restart, delete the ME2 License from "C:\ProgramData\Electronic Arts\EA Services\License" restart again and install ME2 fresh.

You know what? It works now!
Although uninstalling and removing the licence file didn't help, I installed the game from a another Mass Effect 2 game that I have in Origin. I must have acquired that from a Humble Bundle or something. The game recognizes itself as a Digital Deluxe edition when downloading so I can access all the DLCs I think.
