You are referring to the scene where ryder had to die to get out of a situation? If so then they say the same things during the mission regardless of whether or not you romance them. Cora said the same thing when I romanced her and exactly the same thing when I didnt.

I am guessing that by the other two you mean Cora and Vetra? Just like with Peebee, you'll know them when you see them. Peebee's just comes earlier than the others. They also are in little follow up cutscenes. Both are on EOS. Both you will still get I think, if you have done their loyalty missions, but will play out just a little differently if you're already involved with somebody.
You'll know them when you see them if you decide Cora or Vetra are the romance option you want.

The lock in point is different for each character.
I know with Liam it was very obvious- there was a conv abiut getting serious or staying casual. Jaals wasnt so clearly defined because he doesn't do casual. I think the kiss in his room was the lockin point, so maybe for Cora who also doesnt do casual any first kiss moment is her lock in. Something that clearly states romance over flirting/friendship.
I don't really know with Peebee, she starts casual but won't continue unless its serious as I understand her char.
Doing what you propose could be tricky because unlike say ME1 where the sex scene is in the same spot for all, and is the lockin point. This one is not so clearly defined.

Cora gave Scott the "Peebee? Really?" treatment right after Peeb's "I'm not going anywhere" speech. Pretty much all of them the lock-in is the loyalty mission followup cutscene. You probably could postpone it - but you'd have to never open that door on the ship to keep from triggering it.
