That's exactly the reason why you can't access it anymore. You have to "solve the puzzle" in each vault to get the datacore. If you don't, no datacore for you...Wouldn't be much of a puzzle if you could just stroll back in and get it.
Perhaps not as clear as it could be, but that's what it means when SAM says "you may want to search the area before activating the purification field"...meaning there is a puzzle to solve/something to find.

Is that what that means. I just tend to pick them up on the way out when I'm running for my life. They seem to be available then or am I mixing something up here?
