A screenshot of your staircase would really help to see what is going on.
One thing that I have found that affects stair use by sims is if any fencing that was used around either the top and especially the bottom of the stair. So if you have used any fencing then try removing it to see if it clears up the problem. Then try putting it back.
I had a situation with stairs stacked on top of each other so I could go from the first floor to either the basement or 2nd floor. Sims kept getting routing failures to go down and it turned out that it was a fence section that caused it.
Edit: It could also be an object placed under the landing or intruding into the stairway.
And if you are willing and want one of the helpers here to take a look at your build for further troubleshooting, you can load it to the gallery and I or another helper will download and take a look.
