I know there are already rabbithole bookstores in the game (maybe I am thinking of Sims 3 now that I think about it more than Sims 4) but in the small business vein could a sim setup their own activ...

I think you can make a bookstore in a small business with Businesses and Hobbies but you can also create bookstore with the retail system from Get to Work. I'm not sure about the details though, ie selling your own books, etc. Rabbit hole bookstore are in Sims 3, not Sims 4.
Where would you get the inventory to put on sale? You would have to purchase single copies yourself and then mark them up and put them on those sales tables? Can you request copies of the books you have written yourself? i don't remember seeing that option anywhere.
