Yes it does and from the livestream. If you check the box sims that don’t have fame can’t earn fame the ones with fame will continue to get fame. I am not sure where you are getting your information but you can’t get fame if you never had any when it’s turned off. So basically if you want all your sims to have fame you need to leave that option on and run the risk of celebrities invading your towns.
