I fixed this problem. I did two things and should have tested after each but I wasn't thinking because they were sort of related but not entirely.
I had found a Sulani Super Yacht in the gallery when I was trying to fit something in the large empty water lot in Sulani. My intention was to purchase it as a vacation home using LittleMsSams mod More Buyable Venues and New Venue types. Once I did my sims could not take an actual vacation to go there. So that rather ruined that for me. So I decided to turn it into a vacation rental but the lot was designated as a special venue and I couldn't do anything. So I found a mod called Venue changes made by Zerbu and finally was able to change the lot type. At some point during this I had my sim sell it because I wasn't interested in them owning a vacation rental. Once it was done I rented it out and tried to start a social event to throw a party and it was not possible so I was over it and just left it figuring I might use it at some point. Since my sim sold it, I'm not sure this had anything to do with the karaoke problem.
So part two is this, and I feel like this is what was causing the problem. There is a residential rental in Henford-on-Bagley, 13 Nettle Lane, that I had taken my sim to for a vacation once and he met some foxes and befriended them. So I decided to buy that vacation rental as a holiday home. To do this using More Buyable Venues and New Venue types I had to go change the lot type to generic and then I was able to buy it. But it was strange as it was listed as a business and people kept just walking in and stuff and I was like ughhh. So I did some decorating and left and hadn't done or decided what I was going to do with it since.
Something was bugging me about this so I went to Sulani and bulldozed the super yacht. Mainly because of a post I read about game saves and large houses with lots of items. Then I took my sim to the lot in Henford and transferred his money back to the household, sold the lot and then went back in to buy build and changed the lot type back to residential rental, saved and went back to my sim's home. Took him and his wife to Planet Honeypop and they were able to sing duets together again. Took them home and went to my other main household and did the same and they too were successful.
So this seems to have been the "what caused" the problem. Just no clue on the why. I am not going to reinstall those two mods I mentioned as nothing worked out how I wanted it to anyway. But I a going to go in and purchase something using only the means the game gives me and see if the problem reoccurs and I will let you know.
I know you had the problem in one of your saves and if you ever figure it out I would love to know what you find.
I tested the goth family in my save (the family with the issue was playing in a premade house in Ravenwood) that had the issue and it worked fine for them.
Seems for me it's only that one family so far (huge coincidence that I picked that save to test).
Family consists of three Sims, one a ghost. I tried the two non-ghost ones and they still have issues.
Since this thread accumulated a few upvotes and I saw it myself consistently with one family I'm going to leave it on Review. Up to EA to decide what to do.

I think that is a good idea since it affected every household in my save and even new ones created. I'd be happy to share a save that still has everything with the problem intact if they want to pick it apart.

The problem has returned. I am going to remove what mods I had added back and clean out files and repair and I'll post an update once done. I am too tired to mess with it today. Just wanted to post this disappointing update before I got ready for bed.

I cleaned out everything and removed mods once again and reinstalled the game. Been playing clean for days and the singing duets issue has not returned as far as I see. I just put two brothers through university and now that they graduated I am going to retest them and my other households. I know you said that you had never used mods on the save you were able to duplicate the problem in, and that got me thinking. Can you give me details about your household as far as jobs, occult, do they have a small business, own any venue or rental properties. Can't hurt for me to try and make a comparable household and see if anything happens. Outside of that I am going to fully play my households for a while each since some have more varied interests than others and see if I can get the problem to present itself while I am mod free.

I got them and tested in a new game and then I added them to my main save and there is no problem with them doing duets together. Non-ghosts do fine and the husband and wife can even sing duets together. This is all with my game still being mod free since reinstallation. I have some ideas and I am gathering some data and will post back once I feel like I have something solid.

Today I decided to play a new household I had made for testing that has just been sitting there. Still fresh game install that I have not added any mods to. The household could not sing duets together when I took them out for Night on the Town. Both singing skill 3 so should be no problems there. I switched over to my save with your family and the mom and daughter could sing duet together just fine. Switched to my main game save and my two brothers household could not sing duets together.
One thing I will say is when I loaded up the household with the brothers, they were not at home. It showed them to be at the, umm, Starlight Lounge. I think that is the name of it. The karaoke place in San Myshuno that is on the top of a building. Logging in and finding my sims in random places is a new thing and it has been happening a lot. I always put my sims to bed, save, then quit the game or move on to another household. Thought I would mention this and I am going to leave it at that since it is rather off topic and may or may not be pertinent.
Basically the singing duets problem has returned, is not consistent across saves at the same time and is in no way related to mods (that I can see). With that said I do not see what more I could test. It is all above my head now. If there is anything I can do to help expedite a resolution let me know and I'd be happy to oblige.
