Hey, I had the exact same issue. After hours for searching for a fix I just got an idea to rename the settings folder which is in My Documents/BioWare/Mass Effect. I renamed the folder, ME launched correctly. You probably have the same issue - you upgraded your system (I did from Win 7 to Win 10), probably some old settings are incorrect/game directory changed.

I copied my "Save" folder from old to new and works like a charm. Funny that I did not have such issues with ME2 and ME3.
