So like for Augusta, they move the pins to the real traditional positions that the course has on each day of the week of the tournament. I imagine this is the case for other courses as well, but I have pretty much only memorized where pins are at certain days on the Masters, so can't be sure.
Generally speaking, Sunday pin positions are more difficult, and Friday are the easiest. That said, that's not necessarily true for individual holes. While overall Sunday may be more difficult, certain holes may be easier.

Also, same with Tee's, what do the colors mean? And which color is easiest to hardest?
With regard to tee colors, in very general terms:
Black: Tournament
Blue: Low handicap golfer
White: Average amateur
Red: Ladies, youth and the differently abled
