Im still having issues. Normally the “move sims 4 file to desktop” thing works for me but this time the file didnt reset itself. Tried everything else on the list here and still cant get the game to open. Have also just uninstalling/reinstalling the game, repairing, and even restarting the computer after all this. My EA/Sim 4 folder wont respawn either.
@inuyasharules659 The Sims 4 folder inside Documents > Electronic Arts won't respawn until the game loads for the first time, so if it's blocked from loading, it's no surprise that you don't see a new folder.
Are you specifically seeing the already running error, or something else? Please also try setting an exception in your antivirus software, or Controlled Folder Access if you only use Windows Defender, for TS4_DX9_x64.exe, the new executable as of this patch.

Please set an exception inf your antivirus software, or Controlled Folder Access if you're using Windows Defender, for TS4_DX9_x64.exe, the game's new executable. If that doesn't help, please try the other suggestions in the first post on page one.

Im not seeing that new file in windows defender but everything else is set to have access. I even tried turning it off temporarily and that didnt help.

I was having the same issue and none of my EA games would work so I figured it might be due to the timed rewards campaign that is being rolled out, so I change my System Time Setting to Manual and changed the date to tomorrow. I then launched my game, and it started right up. I then closed my game, change the System Time back to Auto and corrected the date and the game still launches. Weird, but it fixed the problem for me.

@inuyasharules659 Again, are you specifically seeing the "Sims 4 is already running" error, or something else?
