Nevermind, I was able to fix it by turning off MSI Afterburner & Rivatuner. For some reason, those applications completely break the game when launching from Steam.

Hey. MSI Afterburner is a tool that you can use to adjust and monitor your graphics card. It also hooks up with a different program called Rivatuner, which when used together lets you have an overlay that shows your computer's temperature, resource usage, clock speed, etc. Rivatuner also lets you set a framerate cap for games, which is useful if there's no ingame options for that.
Something about these two programs causes Mass Effect 3 to not launch for me. It could be because they are "hooking" into the .exe- but I don't know to what extent. If you know for sure that you aren't using those two programs, try checking for other stuff that might be running on your computer.
Although these weren't the problem for me, RGB software like Armory Crate or ICUE might interfere with the game. Again, I didn't have this issue myself, and I personally use ICUE, Razer Synapse, MSI Center (the service, not the application), L-Connect 3 (again the sevice) and Kanali on my system. As you can see, I use plenty of these programs and they didn't affect my game, but it's worth checking anyway.
I guess the main thing to look for would be stuff that might try to hook into or look at the game when it's running. Check your system tray and try closing each program one by one until you figure out what's causing the problem- that's how I figured out MSI Afterburner and Rivatuner were causing the problem for me in my case. Wish you luck bro.
